A Study of Crude Exotoxin of Pseudomonas aeruginosa

Abstract
In recent years the exotoxin of Pseudomonas aeruginosa infection have been observed as an important invasive factors of the toxic substances. So I prepared and isolated the exotoxin from the cultural filtrates of Pseudomonas aeruginosa PA-103 strain, and observed the biologic activity for crude exotoxin.
Strain PA-103 was grown in a dialyzed medium of trypticase soy broth enriched with 1% glycerol and 1 M monosodium glutamate and then immediately after centrifugation,zinc acetate was added to thd supernatant fluid. The mixture was left for a few hour and theh centrifuged to collect the precipitate, which was dissolved in sodium citrate, and then obtained the crude PA-103 strain exotoxin.
When mice lethal toxicity of Pseudomonas aeruginosa PA-103 strains was inoculated into the peritoneal cavity, LD?? was 10??. The supernatant fluid (18 hours) of this strain was showed the 40% low lethal rate on undiluted concentration but LD?? of crude exotoxin was 44.54§¶.
Immunization of rabbit with crude exotoxin resulted in development of antitoxic serum. This antitoxic serum was showed distinctly gel diffusion pattern with Ouchterlony method but precipitate valules was low as 1:16.
Skin responses of rabbit with the crude exotoxin was showed the reddish induration and necrosis etc. 1 Minimum Red Dose (MRD) was below 0.125§¶.
Crude exotoxin of antitoxic neutralization in the skin responses of neutralization test in vitro neutralized the LD?? as 1:16, and in vivo skin neutralization responses was slightly below than this.
